زمینه و اهداف:  لپتوسپیروزیس، از شایع‌ترین بیماری ­های مشترک بین انسان و حیوان در سراسر جهان است که در مناطق گرمسیری، نیمه گرمسیری و معتدل و مرطوب که دارای بارندگی زیاد می­ باشند، بیشتر رخ می­ دهد. تشخیص صحیح و به موقع این بیماری حائز اهمیت می ­باشد. Loa۲۲ یک پروتئین غشای خارجی و در معرض سطح در برخی سرووارهای لپتوسپیرا است که هدف این تحقیق بررسی حضور ژن بیان کننده این پروتئین در سرووارهای باکتری لپتوسپیرا اینتروگانس بود.
مواد و روش کار:  در این تحقیق از ۲۳ سرووار بیماری‌زای لپتوسپیرا و دو سرووار غیر بیماری‌زای لپتوسپیرا استفاده شد. این سرووارها از کلکسیون میکروبی آزمایشگاه رفرانس لپتوسپیرا بخش میکروب شناسی موسسه تحقیقاتی واکسن و سرم سازی رازی کرج در سال ۱۳۹۹ تهیه شدند. پس از استخراج DNA ژنومیک با استفاده از روش استاندارد فنل کلروفرم، ژن loa۲۲ با استفاده از پرایمرهای اختصاصی تکثیر شد.
یافته ها:  نتایج PCR روی ژن loa۲۲ با تولید قطعه‌ای به طول bp ۶۷۱ انجام گردید و مشاهده شد که در همه ۲۳ سرووار بیماری‌زای لپتوسپیرا ژن loa۲۲ وجود دارد، درحالی که این ژن در لپتوسپیراهای غیربیماری‌زای L.biflexa مشاهده نگردید. اختصاصیت پرایمرهای مورد استفاده نیز مورد تایید قرار گرفت.
نتیجه‌گیری:  ژن loa۲۲ یک ژن اختصاصی سرووارهای بیماری­زای لپتوسپیرا است که در سرووارهای ساپروفیت مشاهده نمی­ شود، لذا پیشنهاد می ­شود که از این ژن برای تشخیص سرووارهای بیماری­زای لپتوسپیرا استفاده شود.
