زمینه و اهداف: بیماری مالاریا به عنوان یکی از تهدیدکننده ترین بیماری های عفونی در سطح جهان مطرح می باشد. انتقال انگل از انسان به پشه ناقل توسط گامتوسیت انگل پلاسمودیوم انجام می شود در حالی که این سلول ها در ایجاد علائم بیماری هیچ نقشی ندارند. در حال حاضر در راستای تلاش برای ریشه کنی بیماری در سطح جهانی، کنترل مرحله انتقال انگل از طریق داروهای ضد گامتوسیت به شدت مورد تأکید می باشد. در این تحقیق اثر ضد انگلی رنگ ائوزین B در شرایط برون تنی بر روی مراحل خونی و گامتوسیت پلاسمودیوم مورد مطالعه قرار گرفته است.
مواد و روش کار: ابتدا انگل پلاسمودیوم فالسیپاروم سویه ۳D۷ کشت داده شده و سپس با القای گامتوسیتوژنز، سلول های گامتوسیت تهیه شدند. پس از بررسی مراحل نمو انگل طی دوره کشت انگل با بررسی میکروسکوپی، ائوزین B بر روی انگل های خونی و گامتوسیت تست شده و میزان زنده مانی با استفاده از تست آنزیم لاکتات دهیدروژناز مورد بررسی قرار گرفت. نتایج برای گروه جنسی و غیرجنسی مقایسه گردید.
یافته ها: گامتوسیت ها پس از ۱۲ روز به بلوغ کامل (مرحله V) رسیدند. انگل های غیرجنسی در طول دوره کشت تا روز چهارم افزایش و سپس شروع به کاهش نمودند. نتایج تست آنزیم لاکتات دهیدروژناز حاکی از تاثیر قوی تر ائوزین B بر روی گامتوسیت (IC۵۰=۲۳ nM) نسبت به مرحله غیرجنسی (IC۵۰= ۱۳۳ nM) بود.
نتیجه‌گیری: با توجه به تعداد معدود داروهای ضد گامتوسیت و اهمیت این ترکیبات در کنترل بیماری مالاریا ائوزین B کاندیدای مناسبی برای درمان های ترکیبی علیه گامتوسیت پلاسمودیوم فالسیپاروم می­باشد.
