en تزریق دوز بوستر همچنان در برابر سویه های Ba.4 و Ba.5 موثر است Booster Shot Is Still Effective Against Omicron Ba.4 and Ba.5 Subvariants ویروس شناسی پزشکی Medical Virology نامه به سردبیر Letter to the Editor <div style="text-align: justify;">SARS-CoV-۲ از خانواده ی کرونا ویروس ها است که موجب ایجاد COVID-۱۹ که برای اولین بار در سال ۲۰۱۹ (گروه مطالعاتی Coronaviridae کمیته بین المللی طبقه بندی ویروس ها) در ووهان از چین شناسایی شد می شود. Omicron یکی از واریانت های SARS-CoV-۲ می باشد که اولین بار در ۲۴ نوامبر ۲۰۲۱ از طریق شبکه ژنومیکس آقریقای جنوبی به WHO (بهداشت جهانی) اطلاع داده شد(۱). Omicron ابتدا در بوتسوانا شناسایی شد و سپس در کل دنیا پخش شد و به واریانت غالب تبدیل شد(۲). بعد از BA.۱ (واریانت اصلی Omicron) چندین زیر سویه omicron، مانند BA.۲، BA.۳، BA.۴، و BA.۵ نیز شناسایی شد(۳). زیر سویه های BA.۴ و BA.۵ توسط VTG به عنوان V-۲۲APR-۰۳ و V-۲۲APR-۰۴ طبقه بندی شد. جهش هایی که در پروتیین سنبله BA.۴ هستند عبارتند از: NSP۴: L۴۳۸ (WT، wildtype)؛ S: حذف ۶۹/۷۰، L۴۵۲R، F۴۸۶V، Q۴۹۳ (WT); ORF ۷b: L۱۱F; N: P۱۵۱S و S حذف ۷۰/۶۹. همه ی جهش های  BA.۵ مشابه جهش های BA.۴ هستند و تفاوت هایی که وجود دارد عبارتند از : M: D۳N; ORF ۶: D۶۱ (WT)؛ ORF ۷b: L۱۱ (WT); N: P۱۵۱ (WT); SNP های مترادف: A۲۷۰۳۸G و C۲۷۸۸۹T (۴). برخی از مردم اعتقادشان بر این است که از آنجایی که جهش هایی در زیر سویه های جدید ایجاد شده است پس واکسن ها بی اثر هستند و به همین خاطر از تزریق دوز بوستر امتناع می کنند. این مطالعه با هدف بررسی جهش های Ba.۴ و BA.۵ با تجزیه و تحلیل بیوانفورماتیکی انجام شده است. از آنجایی که برخی از واکسن های مورد استفاده در حال حاضر RBD را هدف قرار می دهند (Receptor Binding Domain). در این تحقیق جهش های RBD مورد بررسی قرار گرفت. نتایج به دست آمده از تجزیه و تحلیل نشان داد که حداقل پنج اپیتوپ با امتیاز بالا مر بوط به سلول B وجود دارد که بدون تغییر باقی مانده است(شکل ۱) (۵). تجزیه و تحلیل اپی توپ های سلول T نشان داد که هنوز تعداد زیادی از اپیتوپ ها بدون تغییر باقی مانده است (بسیاری ازHLA-A، HLA-B و HLA-C). نتایج تحلیل و بررسی های بوجود آمده نشان می دهد که واکسن های مورد استفاده می توانند سیستم ایمنی را علیه زیر سویه های جدید نیز تحریک کنند و موجب تقویت ایمنی سلولی و ایمنی همورال شده و در ممانعت از بستری و مرگ نقش موثری داشته باشند.</div> Severe acute respiratory syndrome coronavirus-2 (SARS‑CoV‑2) is a strain of coronavirus that causes COVID-19 that was first identified in Wuhan of, China, on 2019 (Coronaviridae Study Group of the International Committee on Taxonomy of Viruses, 2020). Omicron variant is one of the SARS-CoV-2 variants that was first informed to WHO (World Health Organization) via South Africa (Network for Genomics Surveillance) on 24 November 2021 (Quarleri et al., 2020). The omicron was first detected in Botswana, and then the variant has spread to become omicron's predominant variant in circulation worldwide (Gowrisankar et al., 2022). Behind the original BA.1 variant (the first dominant omicron variant), several subvariants of omicron, such as BA.2, BA.3, BA.4, and BA.5 (Yao et al., 2022). Omicron subvariants BA.4 and BA.5 were classified as V-22APR-03 and V-22APR-04 by the VTG. BA.4 was designated based on potentially biologically significant mutations in spike. The BA.4 mutations are NSP4: L438 (WT, wild type); S: 69/70 deletion, L452R, F486V, Q493 (WT); ORF 7b: L11F; N: P151S and the S gene 69/70 deletion. The BA.5 has similar mutations/deletions with BA.4 except M: D3N; ORF 6: D61 (WT); ORF 7b: L11 (WT); N: P151 (WT); synonymous SNPs: A27038G, and C27889T (UK health security agency, 2022). Some people believe that the currently used vaccines are ineffective against new subvariants (Ba.4 and BA.5), which is why they do not get vaccinated. This study aimed to investigate the Ba.4 and BA.5 mutations by Bioinformatic analysis. Since some currently used vaccines target the RBD (Receptor Binding Domain) of the S protein of SARS-CoV-2, in this research, the mutations of RBD have been investigated. The results obtained from the IEDB analyzer showed that there are a minimum of five highest score B cell epitopes that remained unchanged (Figure 1). T-Cell epitopes analysis indicated that there are still a lot of unchanged (lots of HLA-A, HLA-B, and HLA-C) and common epitopes (for different MHCI) between the original variant (Wuhan) and BA. 4/5 variants.
