زمینه و اهداف: بروسلوزیس یک بیماری مشترک بین انسان و حیوان می‌باشد و ارزیابی آنتی‌ژن‌های مختلف آن ازنظر پاسخ‌های ایمونولوژیکی و بررسی استراتژی‌های مختلف ایمن‌سازی در طراحی واکسن‌های مؤثر نقشی اساسی دارند. در این تحقیق هدف ما ارزیابی ایمونولوژیکی وکتور یوکاریوتی pVAX1 حامل ژن پروتئین غشای خارجی 31 کیلو دالتونی (Omp31) بروسلا ملی تنسیس بود.

مواد و روش کار: در این مطالعه که در سال 1392 به انجام رسید  توالی ژن omp31 بروسلا ملی تنسیس در پلاسمید pVAX1 بین جایگاه‌های BamHI و XhoI کلون شد. از این پلاسمید برای ایمن‌سازی موش‌های ماده 6-8 هفته‌ای BALB/c (تهیه شده از بخش حیوانات ازمایشگاهی انستیتو پاستور ایران )به‌صورت تزریق داخل ماهیچه‌ای 100 میکروگرم استفاده شد. تزریق یادآور به دو شکل پلاسمیدی یا پروتئینی در گروه‌های جداگانه انجام گرفت. پس از ایمن‌سازی سنجش سطح IFN-γ، IL-4 و IL-12، ایمونوگلوبولین G تام سرمی و IgG1 و IgG2a نسبت به پروتئین نوترکیب Omp31 انجام شد. درنهایت پاسخ ایمنی حفاظت کننده  به دنبال مواجه‌سازی با بروسلا ملی تنسیس 16M مورد ارزیابی قرار گرفت.

یافته‌ها: DNA-واکسن حامل omp31 به همراه یادآور پروتئینی Omp31، میزان بیشتری از IFN-γ، IL-12 و IgG2a را نسبت به گروه‌های دریافت‌کننده DNA-واکسن یا پروتئین نوترکیب تنها تحریک کرده بود. نتایج چالش با سویه بیماری‌زا نیز حاکی ایمنی حفاظت بخش بیشتر در گروه دریافت‌کننده DNA-واکسن و پروتئین بود.

نتیجه‌گیری: بررسی سطح سایتوکین‌ها حاکی از مقدار بالاتر IFN-γ، IL-12 در موش‌هایی بود که با رژیم DNA-واکسن و پروتئین نوترکیب ایمن شده بودند. میزان بالاتر IgG2a سرمی در گروه یادشده به‌موازات پاسخ سایتوکینی نشانگر شکل‌گیری ایمنی سلولی در گروه ذکرشده است که پاسخ ایمنی حفاظت بخش در مقابل سویه بیماری‌زای آن را تأیید می‌نماید.
