زمینه و اهداف:  اشریشیا کلی، فلور طبیعی روده انسان و حیوان است که گاهی باعث بیماری در روده و مجاری ادراری می شود. مطالعه حاضر به بررسی ژن های مقاومت در اشریشیا کلی جدا شده از سرویس های بهداشتی دانشگاه آزاد اسلامی مشهد، ایران و توالی یابی ژنوم در سویه های مقاوم به تتراسایکلین می پردازد.
مواد و روش کار:  این تحقیق مقطعی بر روی ۲۰۰ نمونه جمع آوری شده طی سه ماه انجام شد. نمونه ها در محیط ائوزین متیلن بلو آگار  (EMB)کشت و سپس باکتری ها جدا شدند. ایزوله های اشریشیا کلی با استفاده از واکنش زنجیره ای پلیمراز (PCR)، شناسایی و برای ارزیابی ژن های مقاومت استفاده شدند. ایزوله های اشریشیا کلی، برای تست آنتی بیوگرام به منظور شناسایی سویه های مقاوم استفاده شدند. برای تجزیه و تحلیل آماری الگوی مقاومت آنتی بیوتیکی، از نرم افزار های SPSS-۲۲ و Kolmogorov-Smirnov استفاده شد.
یافته ها:  از ۲۰۰ نمونه، ۴۱ ایزوله به‌عنوان اشریشیا کلی با بالاترین میزان مقاومت به سفوتاکسیم (۷۴/۶۰%) و بیشترین میزان حساسیت به جنتامایسین (۵۸/۷۴%) شناسایی شدند. از بین ۲۰ ایزوله، به ترتیب، ۳۰%، ۲۰% و ۲۵% ژن های blaTEM-۱، blaOXA-۱۰  و blaOXA-۴۸  را داشتند. از بین ۳۸ ایزوله جدا شده مقاوم به تتراسایکلین، سه ایزوله (۷/۸۹%) دارای ژن tetA و دو ایزوله (۵/۲۶% درصد) دارای ژن tetR بودند.
نتیجه‌گیری:  در این تحقیق اکثر نمونه های جمع آوری شده، آلوده به اشریشیا کلی و مقاوم به آنتی‌بیوتیک‌ها بودند. برای کنترل شیوع بیماری عفونی اشریشیا کلی و استفاده خودسرانه از آنتی بیوتیک ها، انجام برخی برنامه های آموزشی و فعالیت های اجتماعی با هدف افزایش آگاهی سلامت توصیه می شود.
