زمینه و اهداف:  بیماری‌های عفونی از اصلی‌ترین دلایل مرگ‌‌ومیر در جهان هستند. استفاده از آنتی‌بیوتیک‌ها برای مقابله با این بیماری‌ها علاوه بر محدودیت‌ها و بروز عوارض جانبی، سبب مقاومت میکروارگانیسم‌ها می‌شود. استفاده از نانوسامانه‌‌های دارورسان راهی موثر برای افزایش پایداری و کاهش میزان مصرف آنتی‌بیوتیک است. هدف این پژوهش بارگذاری داروی سفازولین در نانوسامانه نیوزومی به منظور رهایش کنترل شده آن است.
مواد و روش کار:  این مطالعه تجربی در سال ۹۷ برای بررسی اثر فرمولاسیون بر پایه اسپن ۶۰ ، توئین ۶۰ و کلسترول بر سنتز نانوسامانه نیوزومی بارگذاری شده با آنتی‌بیوتیک سفازولین به روش آب‌رسانی لایه نازک انجام شد. مشخصات نانوسامانه‌های سنتزی و فعالیت ضدباکتریایی آن‌ها مورد بررسی قرار گرفت.
یافته ها:  تصاویر SEM نشان داد، نانوذرات کروی هستند. بازده محصور‌سازی فرمولاسیون اول، دوم و سوم به ترتیب %۳۳، ۱۹/۷% و ۴۰/۷۶% بود. رهایش سفازولین از نانوسامانه اول، دوم و سوم طی ۳۰ روز ۴۸%، ۸۱/۵% و ۶۳% بود. اندازه و پتانسیل زتا نانوسامانه نیوزومی سوم ۱۵۴ نانومتر و ۲۴- میلی‌ولت ارزیابی شد. حداقل غلظت بازدارنده رشد باکتری اشریشیا کلی و استافیلوکوکوس اورئوس به ترتیب ۴ و ۱۵۰ میکروگرم بر میلی‌لیتر است. نانوسامانه با فرمولاسیون سوم اثر ضدباکتریایی معنی‌داری در ۶ روز بر باکتری اشریشیا کلی نشان داد و قطر هاله عدم رشد آن تقریبا ثابت ‌ماند.
نتیجه‌گیری:  فرمولاسیون بهینه نانوذرات نیوزوم شامل اسپن۶۰ (۰/۰۶۰)، توئین ۶۰ (۰/۰۹۰) و کلسترول (۰/۰۴۶) بود. رهایش مداوم و کنترل شده سفازولین از نیوزوم، همراه با افزایش نفوذ دارو سبب کاهش رشد باکتری‌های اشریشیا کلی (ATCC ۹۶۳۷E. coli, ) و استافیلوکوکوس اورئوس (ATCC ۱۲۶۰۰S. aureus, ) می‌شود.
