زمینه و اهداف:  عفونت های جنسی منتقل شده توسط باکتری یکی از مشکلات درمانی و اجتماعی در سراسر جهان است. روش Real-time PCR  یکی از حساس ترین روش های تشخیصی و غربالگری این عفونت ها است. هدف از این مطالعه شناسایی همزمان کلامیدیا تراکوماتیس و مایکوپلاسما ژنیتالیوم با استفاده از TaqMan duplex real-time polymerase chain reaction بود.
مواد و روش کار:  در مطالعه حاضر از DNA استخراج شده از زنان مبتلا به عفونت واژن موجود در بانک مرکز تحقیقات پزشکی و مولکولی دانشگاه علوم پزشکی اراک، اراک، ایران استفاده شد. duplex real-time PCR برای تشخیص کلامیدیا تراکوماتیس و مایکوپلاسماژنیتالیوم در ۱۱۰ نمونه واژینال و اندوسرویکال مورد ارزیابی قرار گرفت. برای تایید عملکرد تشخیصی روش طراحی شده، آزمایش ها با یک کیت تشخیصی تجاری مقایسه شدند.
یافته ها:  اندازه آمپلیکون‌های ۱۳۵ جفت باز OmpA و ۱۴۵ جفت باز MgPa، نشان‌دهنده اجرای درست مراحل طراحی پرایمرها بود. تجزیه و تحلیل پانل STI انسانی هیچ واکنش متقابلی را در روش نشان نداد. حساسیت و ویژگی روش تشخیصی طراحی شده TaqMan duplex Real-time PCR به ترتیب ۹۷% و ۷۵% در مقایسه با کیت تشخیصی تجاری بود.
نتیجه‌گیری:  نتایج نشان داد که تکنیکTaqMan duplex real-time PCR  استفاده شده، اختصاصی و مقرون به صرفه است و می‌تواند جایگزینی عالی برای کیت‌های تجاری برای تشخیص همزمان کلامیدیا تراکوماتیس و مایکوپلاسما ژنیتالیوم باشد. اگر چه محدودیت این روش هزینه بالای آن است، اما این عیب به میزان قابل توجهی با روش مالتی پلکس برطرف شده است.
