جداسازی آمیبهای آزادزی و شناسایی مولکولی آکانتامبا از آبهای راکد کاشان

<span style="font-size:12px;"><span style="font-family:iransans;"><strong>زمینه و هدف:</strong> <em>آکانتامبا</em> شایعترین آمیب آزادزی است که بهوفور در آب، خاک و گردوغبار وجود دارد و عامل آنسفالیت گرانولوماتوز آمیبی است. مطالعه حاضر برای تشخیص آمیب ­های آزادزی و شناسایی ژنوتایپهای <em>آکانتامبا</em> در آبهای راکد شهر کاشان انجام شده است.<br>
<strong>مواد و روش کار:</strong> در این مطالعه توصیفی، ۱۳۸ نمونه آب راکد از مساجد و پارکهای کاشان جمعآوری شد. این نمونهها پس از عبوردادن از فیلتر ۰/۴۵ میکرونی و کشت در آگار غیرمغذی از نظر آمیب آزادزی بررسی شدند. برای شناسایی مولکولی <em>آکانتامبا</em> از واکنش زنجیرههای پلی مراز و پرایمرهای اختصاصی استفاده شد. پس از تکثیر قطعۀ <span dir="LTR">bp</span>۴۹۰ و تعیین توالی نوکلئوتیدی، ژنوتایپ ۱۰ ایزوله <em>آکانتامبا</em> شناسایی شد. <span dir="LTR">pH</span>، دما، فصل نمونهگیری و نتایج در <span dir="LTR">SPSS</span> نسخۀ ۱۶ ثبت و تحلیل شد.<br>
<strong>یافتهها:</strong> ۸۸/۴ درصد از نمونهها آلوده به آمیب آزادزی بودند که با استفاده از<span dir="LTR">PCR </span> ۵۹/۴ درصد آنها بهعنوان <em>آکانتامبا</em> شناسایی و تأیید شدند. از میان نمونههای تأییدشده ۸۰ درصد مربوط به ژنوتایپ <em>آکانتامبا</em> <span dir="LTR">T۴</span> و ۲۰ درصد <span dir="LTR"> T۲</span>بودند. بین فصول نمونهگیری و آلودگی با آمیب آزادزی ارتباط معنیداری مشاهده شد (۰/۰۱ = <em><span dir="LTR">P</span></em>)<span dir="LTR">.</span> بیشترین میزان آلودگی آب راکد به آمیب آزادزی و <em>آکانتامبا</em> در ۷<span dir="LTR">pH</span> مشاهده شد. در این مطالعه ارتباط معنیداری بین <span dir="LTR">pH </span>و دما با آلودگی آمیب آزادزی و <em>آکانتامبا</em> مشاهده نشد.<br>
<strong>نتیجهگیری: </strong>این مطالعه نشان داد که آلودگی آبهای راکد به آمیب آزادزی و <em>آکانتامبا</em> در کاشان بالا است. همچنین ژنوتیپ غالب <em>آکانتامبا</em><span dir="LTR">T۴ </span> است که از سویههای بیماریزا است. با توجه به عوارض خطرناک ناشی از این آمیب، آموزش بهداشت برای افزایش آگاهی در زمینه انتقال و نیز اقدامات بهداشتی برای پیشگیری از آلودگی توصیه میشود. <span dir="LTR"><span style="font-size:9.0pt;"></span></span></span></span><br>

<div style="text-align: justify;"><span style="font-family:times new roman;"><strong>Background and Aims:</strong> <em>Acanthamoeba</em>, the causative agent of granulomatous amebic encephalitis <span dir="RTL">)</span>GAE), is among the most prevalent free-living amoebas (FLA) existing in water, soil and dust. This study was conducted to determine FLA and identify <em>Acanthamoeba </em>genotypes isolated from stagnant water in Kashan, Iran.<br>
<strong>Materials and Methods: </strong>In this descriptive study, 138 stagnant water samples were collected from Kashan mosques and public parks. The samples were filtered (0.45µm) and cultured onto non-nutrient agar for the presence of FLA. <em>Acanthamoeba </em>spp. was identified by polymerase chain reaction (PCR) using specific primers, which amplified a 490 bp fragment. Among ten sequenced isolates of <em>Acanthamoeba</em>, different genotypes were determined by sequence analysis. The parameters such as pH, temperature, sampling season and related results were recorded and analyzed using SPSS16.<br>
<strong>Results: </strong>The rate of FLA was 88.4 %, 59.4% of which were confirmed as <em>Acanthamoeba </em>spp<em>. </em>using PCR method. The rate of <em>Acanthamoeba</em> T4 and T2 genotypes were 80% and 20%, respectively. There was a significant relation between FLA rate and sampling season (<em>P</em>= 0.01). The highest rate of FLA and <em>Acanthamoeba</em> was observed at pH 7. There was no significant relationship between FLA and <em>Acanthamoeba spp</em>. with pH and temperature.<br>
<strong> Conclusions: </strong>The rate of FLA and <em>Acanthamoeba</em> in stagnant water were high in Kashan. The dominant <em>Acanthamoeba</em> genotype (T4) is pathogen. Due to serious amoeba-induced complications, hygienic education is recommended to increase the public awareness on transmission and health/preventive measurements.</span></div>
