بررسی رابطۀ غلظت ویتامین K2 و میزان بیان ژنهای mecA و blaZ در ایزولههای بالینی استافیلوکوکوس اورئوس دارای مقاوم به متیسیلین

<span style="font-size:12px;"><span style="font-family:iransans;"><strong>زمینه و هدف</strong><strong>:</strong> ویتامین <span dir="LTR">K۲</span> به<span dir="LTR"></span>عنوان یکی از ویتامین­های محلول در چربی، می­تواند در برخی موارد آثار مهاری مناسبی بر فعالیت <em>استافیلوکوکوس اورئوس</em> مقاوم به متیسیلین داشته باشد. هدف از این مطالعه تعیین غلظت ویتامین <span dir="LTR">K۲</span> بر میزان بیان ژنهای <em><span dir="LTR">mecA</span></em> و <em><span dir="LTR">blaZ</span></em> در ایزولههای <em>استافیلوکوکوس اورئوس</em> مقاوم به متیسیلین است.<br>
<strong> مواد و روشکار</strong><strong>:</strong> ۷۶ ایزوله بالینی <em>استافیلوکوکوس اورئوس </em>مقاوم به متی­سیلین <span dir="LTR">(MRSA)</span> با تست<span dir="LTR"></span>های فنوتیپی از نمونه<span dir="LTR"></span>های بالینی مختلف (خون، ادرار، زخم و غیره) جداسازی شد. برای تیمار ایزوله<span dir="LTR"></span>های <span dir="LTR">MRSA</span>، از غلظت­های ۱، ۱۰، ۵۰، ۱۰۰، ۲۰۰، ۳۰۰ و ۵۰۰ میکروگرم بر میلی<span dir="LTR"></span>لیتر از ویتامین <span dir="LTR">K2</span> و بازه<span dir="LTR"></span>های مختلف زمانی، استفاده شد. عملکرد ویتامین در زمان<span dir="LTR"></span>های ۲۴، ۴۸ و ۷۲ ساعت گرمخانه<span dir="LTR"></span>گذاری، بررسی شد. برای سنجش کمّی ژن<span dir="LTR"></span>ها از روش <span dir="LTR">Real-time PCR</span> استفاده شد. همچنین به<span dir="LTR"></span>منظور آنالیز نتایج بهدست آمده از نرم افزار <span dir="LTR">REST ۲۰۰۸ V۳</span> و <span dir="LTR">SPSS V۱۶</span> استفاده شد.<br>
<strong>یافته ها<em>:</em></strong> میزان بیان ژن های <em><span dir="LTR">blaZ</span></em> و <em><span dir="LTR">mecA</span></em> تنها در غلظت ۵۰۰ میکروگرم بر میلی لیتر از ویتامین 2<strong><span dir="LTR">K</span></strong> <span dir="RTL">کاهش داشت</span>. علاوه بر این، زمان<strong><span dir="LTR"></span></strong>های گرمخانه گذاری ۴۸ و ۷۲ ساعت بهترین اثر عملکردی را از نظر تاثیر ویتامین بر ژن<strong><span dir="LTR"></span></strong>های مورد مطالعه داشت. در ایزوله های جدا شده از نمونه های بالینی زخم و ادرار کاهش بیان ژنهای <em><span dir="LTR">mecA</span></em> و <em><span dir="LTR">blaZ</span></em> بیشترین مقدار را به خود اختصاص داد. همچنین، ارتباط معنی داری بین زمان گرمخانه گذاری و نوع نمونه بالینی بر کاهش بیان ژن های مورد نظر، مشاهده شد (۰/۰۱> p) و (۰/۰۵> p).<br>
<strong>نتیجه گیری<em>:</em></strong> <span dir="RTL">ویتامین </span>K۲<span dir="RTL"> می</span><strong></strong><span dir="RTL">تواند نقش مهمی در کنترل سویه</span><strong></strong><span dir="RTL">های استافیلوکوک اورئوس مقاوم به متی</span><strong></strong><span dir="RTL">سیلین داشته باشد.</span></span></span>
<span style="font-family:times new roman;"><strong>Background and Aims:</strong> As one of the fat-soluble vitamins, vitamin K, can in some cases have intrinsic inhibitory effects on the activity of methicillin-resistant <em>Staphylococcus aureus</em>. The aim of this study, was to determine the effect of vitamin K2 on the expression of <em>mecA</em> and <em>blaZ</em> genes in Multi Drug Resistant Methicilin Resistance <em>S. aureus</em> isolates. <strong><span dir="RTL"></span></strong><br>
<strong>Materials and Methods: </strong>76 clinical isolates of methicillin-resistant <em>S.aureus</em> were isolated by phenotypic tests. For treatment of isolates, concentrations of 1, 10, 50, 100, 200, 300 and 500 μg / ml of vitamin K2 and different time intervals were used. The q-PCR method was used to quantitatively evaluate the genes. It was also used to analyze the results of REST 2008 V3 and SPSS 16 software.<br>
<strong>Results: </strong>The expression of <em>blaZ</em> and <em>mecA</em> genes was reduced only at a concentration of 500 μg / ml of vitamin K2. Additionally, the 48 and 72 hours incubation times had the best effect on the effect of vitamins on the genes studied. In the isolates from clinical specimens, ulcers and urine reduced the expression of <em>mecA</em> and <em>blaZ</em> genes. Also, there was a significant relationship between incubation time and clinical specimen type on decreasing the expression of the desired genes (<em>P</em>≤0.01, <em>P</em>≤0.05).<br>
<strong>Conclusions: </strong>In accordance to the results of this study, the use of vitamin K2 can play an important role in the control of methicillin-resistant Staphylococcus aureus strains.</span><br>
