زمینه و هدف: استافیلوکوکوس اورئوس مقاوم به متی­سیلین شایع­ترین عامل باکتریائی عفونت زخم پای بیماران دیابتی (DFI) است. فاکتورهای بیماری ­زایی این باکتری شدت درجۀ عفونت زخم را افزایش می­دهد و درصورت درمان نشدن به ­موقع منجر به قطع عضو اندام ­های تحتانی و مرگ بیماران مبتلا می­شود. هدف این پژوهش، جداسازی باکتری استافیلوکوکوس اورئوس مقاوم به متی­سیلین حامل ژن mecA  و شناسایی ۴ ژن ویرولانس hla ، lukED، sei و hlg از جدایه­ های عفونت پای بیماران دیابتی به­ منظور ارزیابی نقش آن­ ها در میزان شدت عفونت است.
مواد و روش کار: از ترشحات چرک ۳۰ بیمار مبتلا به عفونت پای دیابتی در استان مازندران نمونه­ گیری شد و استافیلوکوکوس اورئوس براساس ویژگی ­های کشت و تست­ های بیوشیمیائی خالص ­سازی شد. ارزیابی مولکولی جدایه­ ها پس از استخراج DNA، با ۵ جفت پرایمر اختصاصی ژن­های مقصود به روش PCR صورت گرفت.
یافته‌ها: از مجموع ۳۰ بیمار ۱۴ ایزوله (۴۶/۶%) استافیلوکوکوس اورئوس جداسازی و شناسایی شد. هر ۱۴ نمونه حامل ژن mecA  بودند و فراوانی ژن­های hla  ، lukED ، sei و hlg در جدایه­ها به­ ترتیب ۱۰۰%، ۱۰۰%، ۷۱/۴% و ۶۴/۲% به ­دست آمد.
نتیجه‌گیری: شناسایی این ۴ ژن به همراه ژن mecA در استافیلوکوکوس اورئوس جداسازی شده از زخم پای دیابتی می­تواند مارکر ژنتیکی مؤثر و قابل اعتمادی در تشخیص درجه­ بندی عفونت پای بیماران دیابتی باشد. تشخیص عامل عفونت با روش PCR و درمان آنتی­بیوتیکی مناسب و به­ موقع، در جلوگیری از قطع عضو بیماران دیابتی ضروری است.
